Firstly, two types of live cell fluorescentimaging techniques -fluorescence recovery after photobleaching (FRAP) andfluorescence resonance energy transfer (FRET) were developed in livingcyanobacteria cells using laser scanning confocal microscope.
本論文主要分三部分:一是利用激光掃描共聚焦顯微鏡在藍(lán)藻細(xì)胞內(nèi)建立兩種活體內(nèi)熒光成像技術(shù)平臺:熒光漂白恢復(fù)技術(shù)(fluorescence recovery afterphotobleaching, FRAP)和熒光共振能量轉(zhuǎn)移技術(shù)(fluorescence resonance energytransfer, FRET);二是利用FRAP技術(shù)研究與藍(lán)藻狀態(tài)轉(zhuǎn)換有關(guān)的藻膽體的動態(tài)擴(kuò)散及其驅(qū)動力和調(diào)控因素;三是利用活體內(nèi)FRET技術(shù)初步探討了藍(lán)藻NAD(P)H脫氫酶復(fù)合物的亞基之間的距離及相互作用。
